Pada tahun 1947, syarikat penerbangan yang baru dibentuk memulakan operasi berjadual dengan Armada Airspeed Konsul yang tunggal, [175] menghubungkan Lapangan Terbang Singapura-Kallang dengan Kuala Lumpur dan Pulau Pinang, dan Kuala Lumpur dengan Kota Bharu dan Kuantan. [176] Menjelang tahun 1948, rangkaian laluan domestik terdiri daripada Ipoh, Kuala Lumpur, Pulau Pinang, Singapura, Kota Bharu dan Kuantan, sedangkan penerbangan antarabangsa ke Batavia (kini Jakarta), Bangkok, Medan, Saigon (Kini Bandaraya Ho Chi Minh) dan Palembang juga layani. Berpusat di Lapangan Terbang Antarabangsa Subang, Malaysian Airlines System Berhad (MAS) dibentuk oleh kerajaan Malaysia pada April 1971 untuk menggantikan MSA, memulakan operasi pada Tarikh 1 Oktober 1972, sehari selepas tarikh tersebut, Syarikat Penerbangan MSA menjadi tidak berfungsi kerana perpecahannya antara MAS dan Singapore Airlines.
